چکیده

Colony-stimulating factors (CSFs) are a family of hematopoietic growth factors required for the proliferation and differentiation of hematopoietic progenitor cells [I, 2]. In the human system, purification to homogeneity and biochemical characterization has only been reported for macrophage-active CSF (CSF-l) [3]. However, there are many reports about highly purified human granulocyte-macrophage CSFs (e.g. [4-7]), but not about pluripotent human CSF. Assays are available to detect human clonogenic precursors that give rise to cells of the erythroid, granulocytic, megakaryocytic, macrophage, colony-forming unit granulocytes, erythrocytes, macrophages, and megakaryocytes, CFU-GEMM, and possibly lymphoid lineages [8-10]. CSFs with activities on these pluripotential progenitor cells (pluripotent CSF) are produced by mitogenor antigen-activated T-lymphocytes [11] and by human tumorcell lines [12] or HTLV-transformed lymphoid cells [13]. We report in this paper the purification to homogeneity and biochemical charac-

Colony stimulating factors (CSF) 1 a r e a class of hemopoietic regulatory glycoproteins required for survival, proliferation, and differentiation of hemopoietic stem and progenitor cells in vitro, and probably in vivo (1, 2). Many efforts have been undertaken to characterize the biochemical and biological properties of these factors. In the routine system, CSF stimulating the growth of progeni...

Proliferative effects of a recombinant human granulocyte colony-stimulating factor (rG-CSF) on highly enriched hematopoietic progenitor cells.

Human multipotential and committed hematopoietic progenitor cells require the presence of specific glycoproteins, termed "colony-stimulating factors" (CSFs) for survival, clonal proliferation, and differentiation. Recently, a pluripotent G-CSF, constitutively produced by the human bladder carcinoma line 5637, has been purified to apparent homogeneity [11 and molecularly cloned with the compleme...

Efficient Process Development of Recombinant Human Granulocyte Colony-Stimulating Factor (rh-GCSF) Production in Escherichia coli

Background: The protein hormone granulocyte colony-stimulating factor (GCSF) stimulates the production of white blood cells and plays an important role in medical treatment of cancer patients. Methods: An efficient process was developed for heterologous expression of human GCSF in E. coli BL21 (DE3). The feeding rate was adjusted to achieve the maximum attainable specific growth rate under crit...
